特徴

• The CD40 antigen, which contains four homologous repeating cysteine-rich extracellular domains, is a single chain glycoprotein.
• It is known to be a member of the tumor necrosis factor/nerve growth factor superfamily and shows a significant homology to the Hodgkin’s disease-associated antigen, CD30.
• CD40 is present on all B cells except plasma cells.
• It is also found on some epithelial cells, carcinomas and lymphoid dendritic cells.
• CD40 is found on normal and neoplastic B cells, strongly expressed by interdigitating cells (IDC), basal epithelial cells, and carcinomas, and also present on macrophages, some endothelial cells, and follicular dendritic cells.
• It is a pan-B marker, absent only from terminally differentiated plasma cells.
• CD40 receptor is expressed on antigen-presenting cells such as B cells, macrophages, dendritic cells and thymic epithelial cells.
• CD40 mediates stimulation of cell growth, differentiation and death.
• It also plays a role in regulating transcription in B cells.
• CD40 is predicted to function in the process of B cell selection in the germinal center.
• Studies demonstrated that it induces strong homotypic adhesions in resting B cells and, together with Interleukin-4 (IL-4) maintains the cell cycle of B blasts and promotes the switch to IgE secretion.
• Purified B cells cultured in the CD40 system in the presence of IL-10 differentiate into plasma cells and secrete large amounts of immunoglobulins.
• The ligand for CD40 is a membrane glycoprotein on activated T cells that induces B cell proliferation and immunoglobulin secretion.
• Applications:Suitable for use in Fluorescence Microscopy, Flow Cytometry and Functional study of B-cell subpopulations.
• Also suitable for the study of B-cell proliferation, B-cell growth and activation factors.
• Other applications not tested.
• Recommended Dilution:Fluorescent Microscopy/Flow Cytometry: 2ug/5x10e5 cells/test
• Activation Assays: 10ng/ml

• Hybridoma:Hybridization of NS1 myeloma cells with spleen cells from Balb/c mice.
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
• Reconstitute with sterile ddH2O.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
