This is a hematopoietic differentiation antigen (also named SSEA-1, stage-specific embryonic antigen) expressed on most terminally differentiated myeloid cells including granulocytes, monocytes, eosinophils, mast cells, macrophages and Langerhans cells.On progenitor cells CD15 is coexpressed by 0.25 to 1% of bone marrow CD34+ cells.(2) In the peripheral blood growth factor mobilized CD34+ cells expressed CD15
(0.5 to 3.8% positive) in the native state and after neuraminidase exposure (up to 24% positive).(2)
On the umbilical cord blood cells 80H5 stains more of 15% of CD34-bright cells and more of 30% after neuraminidase treatment.(2)
CD15 plays a role in mediating phagocytosis, bactericidal activity, and chemotaxis.
It is present on >95% of granulocytes including neutrophils and eosinophils and to a lesser degree on monocytes.
In addition, CD15 is expressed in Reed-Sternberg cells and some epithelial cells.
CD15 antibody is very useful in the identification of Hodgkin’s disease.
CD15 is occasionally expressed in large cell lymphomas of both B and T phenotypes which otherwise have a quite distinct histological appearance.
This antibody has been assigned to the CD15 cluster of differentiation at the Vth International Workshop on Human Leukocyte Differentiation Antigens in Boston (1993).
Applications:May be used for enumerating mature granulocytes in peripheral blood in Flow Cytometry.
Other applications not tested.
Recommended Dilution:Flow Cytometry: 1ug to stain 1x10e6 cells
Optimal dilution determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
