Prohibitin is an evolutionarily conserved protein with homologues found in yeast to man.
It is located in the inner membrane of mitochondria.
PROHIBITIN APPEARS TO BE A VERY RELIABLE MARKER OF MITOCHONDRIA.
Shows antiproliferative activity and has been proposed to play a role in normal cell cycle regulation, replicative senescence, cellular immortalization, and tumor suppression.
Applications:Suitable for use in Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry.
Other applications have not been tested.
Recommended Dilution:Immunoprecipitation: 2ug/mg protein lysate., (Protein G)
Native and denatured
Western Blot: 0.5-1ug/ml for 2 hours at RT.
Immunohistochemistry (formalin/paraffin): 2-4ug/ml for 30 minutes at RT.
Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.
Optimal dilutions to be determined by the researcher.
Positive Control:MAD109, or PC12 cells, heart
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
