特徴

• Expression of p21WAF1 is induced by wild type, but not mutant, p53 suppressor protein.
• The p21WAF1 protein binds to cyclin/CDK complexes and inhibits their kinase activity thereby stopping cell cycle progression.
• Meanwhile, the p21WAF1 protein also binds to PCNA (proliferating cell nuclear antigen) and blocks DNA replication but not the DNA repair process.
• Evidently, inactivation of p53 disrupts the p53-dependent expression of p21WAF1.
• p21WAF1is a tumor suppressor involved in the pathogenesis of a variety of malignancies.
• Applications:Suitable for use in Immunofluorescence, Immunoprecipitation and Immunohistology.
• Not suitable for Flow Cytometry or Western Blot.
• Other applications have not been tested.
• Recommended Dilution:Immunoprecipitation (native and denatured): 2ug/ml protein lysate.
• Co-precipitates p21 proteins.
• Immunohistochemistry (formalin and paraffin): 1:25-1:50 for 60 minutes at RT.
• Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.
• Optimal dilutions to be determined by the researcher.
• Positive Control:Ls174T or Raji cells and colon carcinoma.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
