Laminins are large heterotrimeric, noncollagenous glycoproteins composed of alpha, beta and gamma chains.
They are ubiquitously present in basement membrane (BM) along with entactin/nidogen (EN), collagen type IV (CIV) and large heparan sulfate proteoglycan (HSPG).
All interact specifically with each other to form a continuous and regular BM.
Alterations of BM integrity, from local discontinuities up to complete loss, are described in many types of human and animal epithelial neoplasms.
Applications:Suitable for use in Immunofluorescence, Immunoprecipitation and Immunohistochemistry.
Other applications have not been tested.
Recommended Dilution:Immunoprecipitation (Native and denatured): 2ug/mg protein lysate using Protein G.
Immunohistochemistry (Acetone-fixed, frozen and acid-alcohol-fixed, paraffin-embedded): 2ug/ml for 30 minutes at RT.
Staining of acid-alcohol fixed tissues requires antigen unmasking with 15,000U/ml of bovine testicular hyaluronidase in PBS, pH 7.4 for 30 minutes at 37°C.
Optimal dilutions to be determined by the researcher.
Positive Control:LS174T cells, normal colon or colon carcinoma.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
