cdc25C (Mr 56 and 59kD), in asynchronous cells, is a dual specificity protein phosphatase that controls entry of cells into mitosis by dephosphorylating and activating the Cdc2/cyclin B complex and is regulated at both the transcriptional and post-translational levels.
Expression of cdc25C is restricted to late S/G2, effected by a transcriptional repressor that binds the CDE (cell cycle-dependent element) in the basal promoter.
cdc25C is also regulated via phosphorylation of Ser216 by C-TAK1 (cdc25c associated protein kinase) which mediates interaction with the 14-3-3 family, maintaining cdc25C “inactive”.
Phosphorylated Ser216 is detectable only throughout interphase.
Cells expressing cdc25C mutants that cannot be phosphorylated at Ser216 exhibit perturbed mitotic timing and escape the G2 checkpoint arrest induced by radiation induce DNA damage.
Applications:Suitable for use in Immunofluorescence, Immunoprecipitation, and Western Blot.
Not suitable for Immunohistochemistry.
Other applications have not been tested.
Recommended Dilution:Immunoprecipitation: Use Protein G; 2ug antibody/mg protein lysate
Western Blot: 1-2ug/ml for 2 hours at room temperature.
Optimal dilutions to be determined by the researcher.

Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
