RAD50, a member of the RAD52 epistasis group, is believed to play an important role in the nucleolytic processing of DNA double-stranded breaks.
Studies in Saccharomyces cerevisiae show that mutation in the RAD52 family of proteins causes varying degrees of mitotic and meiotic recombination, both of which are initiated via DNA double-stranded breakage.
Rad50 forms a complex with Mre11, another RAD52 family member, and p95 Nibrin, a product of the mutated gene associated with Nijmegen breakage syndrome.
This complex possesses both endonuclease activity and a 3' and 5' exonuclease activity.
RAD50 is indispensable for non-homologous DNA end joining and the maintenance of telomere length.
Applications:Suitable for use in Western Blot and Immunoprecipitation.
Other applications have not been tested.
Recommended Dilution:Western Blot: 0.5-2ug/ml detects RAD50 in a HeLa nuclear lysate.
HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with R0450 (0.5ug/ml).
Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Optimal dilutions to be determined by the researcher.
Positive Control Included:H1838: HeLa nuclear extract.
Add an equal volume of Laemmli reducing sample buffer to 5-10ul of extract and boil for 5 minutes to reduce the preparation.
Load 20ug of reduced lysate per lane for minigels.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
