特徴

• RAD50, a member of the RAD52 epistasis group, is believed to play an important role in the nucleolytic processing of DNA double-stranded breaks.
• Studies in Saccharomyces cerevisiae show that mutation in the RAD52 family of proteins causes varying degrees of mitotic and meiotic recombination, both of which are initiated via DNA double-stranded breakage.
• Rad50 forms a complex with Mre11, another RAD52 family member, and p95 Nibrin, a product of the mutated gene associated with Nijmegen breakage syndrome.
• This complex possesses both endonuclease activity and a 3' and 5' exonuclease activity.
• RAD50 is indispensable for non-homologous DNA end joining and the maintenance of telomere length.
• Applications:Suitable for use in Western Blot and Immunoprecipitation.
• Other applications have not been tested.
• Recommended Dilution:Western Blot: 0.5-2ug/ml detects RAD50 in a HeLa nuclear lysate.
• HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with R0450 (0.5ug/ml).
• Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
• Optimal dilutions to be determined by the researcher.
• Positive Control Included:H1838: HeLa nuclear extract.
• Add an equal volume of Laemmli reducing sample buffer to 5-10ul of extract and boil for 5 minutes to reduce the preparation.
• Load 20ug of reduced lysate per lane for minigels.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
