Parathyroid hormone (PTH), or parathormone, is secreted by the parathyroid glands as a polypeptide containing 84 amino acids.
It acts to increase the concentration of calcium (Ca2+) in the blood, whereas calcitonin (a hormone produced by the parafollicular cells (C cells) of the thyroid gland) acts to decrease calcium concentration.
PTH acts to increase the concentration of calcium in the blood by acting upon parathyroid hormone receptor in three parts of the body.
PTH half-life is approximately 4 minutes.
PTH enhances the release of calcium from the large reservoir contained in the bones.
Bone resorption is the normal destruction of bone by osteoclasts, which are indirectly stimulated by PTH.
Stimulation is indirect since osteoclasts do not have a receptor for PTH; rather, PTH binds to osteoblasts, the cells responsible for creating bone.
Binding stimulates osteoblasts to increase their expression of RANKL, which can bind to osteoclast precursors containing RANK, a receptor for RANKL.
The binding of RANKL to RANK stimulates these precursors to fuse, forming new osteoclasts which ultimately enhances the resorption of bone.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications have not been tested.
Recommended Dilutions:Western Blot: 0.5-2ug/ml.
Detects PTH/PTHrP receptor from RIPA lysates of 3T3-L1 pre-adipose fibroblasts.
3T3-L1 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with P3109-50.
Proteins were visualized using an IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system.
Immunohistochemistry: Detects PTH/PTHrP receptor in human kidney, vascular smooth muscle and ovary sections.
Receptor was also detected in porcine oocytes.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
