Vinculin, a cytoskeletal protein found in adherens type cell-cell and cell-extracellular matrix junctions, displays considerable heterogeneity.
At least three 117kD and two 130kD isoforms (muscle specific metavinculin) have been detected.
Vinculin binds talin and a-actinin in adhesion plaques and may play either a structural or a regulatory role in these structures.
Applications:Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-1ug/ml detects Vinculin in RIPA lysates of human A431 cells; previous lots detected Vinculin in RIPA lysates from chicken embryonic fibroblasts (CEF) and mouse 3T3 fibroblasts.
Non-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with V2122-50 (0.5ug/ml).
Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunoprecipitation: 4ug immunoprecipitates Vinculin from mouse 3T3/A31 and human A431 RIPA lysates.
Immunohistochemistry and Immunocytochemistry (frozen and formalin fixed sections): 10ug/ml will stain Vinculin in Ref-52 cells fixed with 4% paraformaldehyde and permeabilized with 0.25% Triton X-100.
Optimal dilutions to be determined by the researcher.
Positive Antigen Control (Included):A0001-50, Non-stimulated A431 cell lysate.
Use 20ug per lane for minigels.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
