• Gab2 (Grb2-associated binder) is the latest member of a new family of multi-functional adapter molecules containing a Pleckstrin Homology domain, defined initially by the cloning of Gab1.
• Gab2 is broadly expressed as a 90-100kD protein.
• It contains two putative SH3 binding motifs and is tyrosine phosphorylated in response to a range of cytokine, growth factor, and antigen stimulation.
• Gab2 associates with SHP2, the p85 subunit of PI3K, Grb2, and SHC demonstrating its involvement in multiple signaling pathways.
• Gab 2 transfection experiments show that Gab2, like Gab1, stimulates Erk kinase activity following EGF-stimulation or co-transfection with activated Ras.
• By contrast, while Gab1 increases activated Ras or EGF-mediated transactivation of Ekl-1 responsive genes, Gab2 has an inhibitory effect.
• Thus, Gab2 may function as a negative regulator of cytoplasmic signaling to the nucleus.
• Applications:Suitable for use in Western Blot.
• Not recommended for Immunoprecipitation.
• Other applications not tested.
• Recommended Dilution:Western Blot: 0.5-2mg/ml.
• Detects GRB2 (23–25kD) in RIPA lysates of non-stimulated human A431 carcinoma cells.
• Previous lots detected GRB2 in RIPA lysates of mouse 3T3 fibroblasts, rat L6 myoblasts, chinese hamster ovary cells and bovine brain extract.
• Optimal dilutions to be determined by the researcher.
