特徴

• Expression of p21WAF1 is induced by wild type, but not mutant, p53 suppressor protein.
• The p21WAF1 protein binds to cyclin/CDK complexes and inhibits their kinase activity thereby stopping cell cycle progression.
• Meanwhile, the p21WAF1 protein also binds to PCNA (proliferating cell nuclear antigen) and blocks DNA replication but not the DNA repair process.
• Evidently, inactivation of p53 disrupts the p53-dependent expression of p21WAF1.
• p21WAF1is a tumor suppressor involved in the pathogenesis of a variety of malignancies.

• Recommended Dilution:Immunoprecipitation: 4ug immunoprecipitates Cip1 from 500ug of HeLa nuclear extract.
• Western Blot: 0.5–2ug/ml detects WAF1, p21 in 20ug of HeLa nuclear extract.
• HeLa nuclear extract was resolved by SDS PAGE, transferred to nitrocellulose, and probed with anti-WAF1, p21 (0.5ug/ml).
• Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
• Positive Control:HeLa nuclear extract.
• Use 20ug per lane for minigels.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
