Apoptosis or programmed-cell death is a physiological process essential for the normal development and maintenance of homeostasis in many organisms.
This “cellular suicide” can be mediated by the Fas antigen (CD95, APO1), a cell-surface glycoprotein, 40-50kD, that belongs to the nerve growth factor/tumor necrosis factor (TNF) receptor family.
Mice carrying the lymphoproliferation (lpr) mutation, which develop lymphoadenopathy and suffer from a systematic lupus erythematosus-like autoimmune disease, have defects in the Fas gene.
Fas is expressed in several tissues including blood, where it expression is upregulated on activated T and B lymphocytes and natural killer cells.
Binding of the Fas antigen by agonistic anti-Fas antibody has been shown to induce apoptosis.
Applications:Suitable for use in Immunocytochemistry, Flow Cytometry and Western Blot.
Suitable for use in inducing apoptosis.
Other applications not tested.
Recommended Dilutions:Immunocytochemistry: 5-10ug/ml detected Fas on HeLa cells fixed with 4% formalin/2% acetic acid.
Flow Cytometry: 20ug/ml (see ref #s 1 and 2).
Western Blot: 0.5-2ug/ml was used for detection of Fas in Raji cell lysate.
Apoptosis analysis: 15-20ug/ml maximally induced apoptosis of human Jurkat cells with 80–90% mortality after 24 hours of treatment.
Optimal dilutions to be determined by the researcher.
Biological Activity:This antibody demonstrates cytolytic activity on human cells that express Fas.
Murine WR19L cells and L929 cells transfected with cDNA encoding human Fas undergo apoptosis in response to this antibody.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Size:50ug
Host:mouse
Source Antibody:human
Grade:Affinity Purified
Purity:Purified by immunoaffinity chromatography.
Form:Supplied as a liquid in PBS, pH 7.2, 50% glycerol.
Specificity:Recognizes the human cell surface antigen Fas (~43kD) expressed in various human cells, including myeloid cells, T lympho-blastoid cells and diploid fibroblasts. This antibody does not recognize TNF and does not crossreact with mouse Fas.
