特徴

• Cortactin is present as a p80 and p85kD protein in many cell types.
• The protein was first identified as being a major substrate of the tyrosine kinase pp60SRC.
• The main structural feature of the protein is that at the N-terminus there are five and a half 37-amino acid repeats, these appear to be responsible for the actin-binding.
• This repeat region occupies about a third of the protein.
• The repeat region is followed by a helical region and then a proline rich region and finally a SH3 domain.
• Applications:Suitable for use in Western Blot, Immunoprecipation and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilutions:Western Blot: 0.5-2ug/ml detects Cortactin (p80/85) in mouse 3T3/A31 cell lysates.
• Detects Cortactin in chicken fibroblast src-transformed cell lysate.
• 3T3/A31 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C7901-80.
• Proteins were visualized using an IgG (HRP) Gt x Mo and a chemiluminescence detection system.
• Immunoprecipitation: Use 5ug for ~500ug of cell lysate.
• Immunocytochemistry: 10ug/ml
• Optimal dilutions to be determined by the researcher.
• Hybridoma:Sp2/0 myeloma x AJ mouse splenocytes
• Included Positive Antigen Control:F4209: 3T3/A31 cell lysate.
• Add 2.5ul 2-mercaptoethanol/100ul of lysate.
• Boil for 5 minutes to reduce the preparation.
• Load 20ug of reduced lysate per lane.
• Note: PC-12 cell lysate, HeLa whole cell lysate or A431 whole cell lysate can also be used as a positive control.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
