The proto-oncogene c-myb was first encountered in chickens as a transduced retroviral oncogene, v-myb, which causes a myeloid leukemia and transforms myeloid cells in culture.
Mutations affecting c-myb have since been implicated in the tumors of both humans and mice.
The product of c-myb (Myb) is a transcription factor that binds a specific DNA sequence.
The protein is divided into at least three discrete functional domains: one for binding to DNA, a second for activation of transcription, and a third domain that governs the biochemical activity of the protein.
It is generally believed that Myb proteins, including MybA and MybB, two additional vertebrate Myb proteins that are related to c-myb, play roles in the cell division cycle.
c-myb and MybB have been implicated in the G1/S transition, whereas MybA is more likely to be involved in cellular differentiaton.
Myb-like proteins with related DNA-binding domains have also been found in yeast and plants.
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilution:Western Blot: 0.5–2ug/ml detects Myb in 20ug of RIPA lysate from Jurkat cells.
Jurkat cell lysate was resolved by electro-phoresis, transferred to nitrocellulose and probed with anti-Myb (0.5ug/ml).
Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunoprecipitation: 4ug of a previous lot immunoprecipitated Myb from Jurkat cell lysate.
Optimal dilutions to be determined by the researcher.
Included Positive Antigen Control: Jurkat cell lysate.
Add 2.5ul of 2- mercaptoethanol/100uL of lysate and boil for 5 minutes to reduce the preparation.
Load 20ug of reduced lysate per lane for minigel.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
