CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180kD-220kD.
It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids.
CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues.
The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains.
Several studies on the function of CD45 in leukocytes support a crucial role of CD45 in the regulation of leukocyte activation, development, and function.
In these processes, distinct CD45 isoforms appear to play differential functions.
The diversity of the structure, size, and carbohydrate content of different CD45 isoforms is cell-type dependent and developmentally regulated.
This structural heterogeneity is mainly the result of the alternative splicing of a single CD45 gene.
Applications:Suitable for use in ELISA.
Other applications not tested.
Recommended Dilution:ELISA: 1:1000
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
仕様
Size:800ug
Host:mouse
Source Antibody:human
Grade:Highly Purified
Purity:Murine ascites purified by column chromatography.
Form:Supplied as a liquid in PBS buffer, pH 7.5, 0.08% sodium azide.
Specificity:Recognizes the 220kD MW component of the leukocyte common antigen complex expressed on some CD4+ T lymphocytes, B lymphocytes and CD8+ lymphocytes. The CD45R+/CD4+ lymphocytes functionally exert suppressor/inducer activity in in vitro assay systems.
