Hepatocyte Plating Medium (HPM) is formulated to aid in the attachment of fresh or cryopreserved hepatocytes to Collagen I, liver biomatrix or in co-culture with human liver stromal cells. HPM is also used for the recovery and plating of human liver stromal cells. HPM is a DMEM base medium without phenyl red and includes stabilized L-glutamine, HEPES and non-essential amino acids.
After HPM Serum-Free Supplements are added to the basal medium and filtered to ensure sterility, the medium is complete and ready to use for plating human hepatocytes and human liver stromal cells. Hepatocyte Plating Medium is complete so you don’t need to source additional components. All supplements required for making this medium complete come separate in frozen vials in the supplement pack
Components:a. Hepatocyte Plating basal medium (250ml bottle), stored at 4°C

Preparing Cryopreserved Hepatocyte Plating Medium:1. Warm the basal medium to room temperature2. Thaw the contents of the frozen supplement packa. Aseptically add the supplements to the basal medium3. Filter through 0.22um filter unit4. Store the complete medium at 4°C in the dark until ready for use
5. Warm the complete medium to room temperature immediately before use
a. Do not leave the complete medium in a water-bath for longer than 5 minutes as this may result in degradation of the medium componentsNote: Degradation of some components is toxic to cells 6. Use complete medium within two weeks
Storage and Stability:Store the complete medium at 4°C in the dark until ready for use.
