Hepatocyte Differentiation Medium contains glutamine and supplemental factors but no antibiotics. After the components of the Hepatocyte Differentiation Serum-Free Supplement Pack are added to the basal medium and filtered to ensure sterility, the medium is complete and ready to use for differentiation of human stem/progenitor cells of the Endodermal lineage (liver, pancreas, lung, intestine) into hepatocytes. Hepatocyte Differentiation Medium is designed to differentiate stem/progenitor cells of the endodermal lineage into hepatocytes within a few days when combined with our Liver Biomatrix.
Components:Includes Hepatocyte Differentiation Basal Media and a supplement pack containing the following components:1. Hepatocyte Differentiation Supplement (contains glutamine)2. EGF3. HGF4. Dexamethasone
Preparing Hepatocyte Differentiation Medium:1. Warm the basal medium to room temperature2. Thaw contents in the Serum-Free Supplementa. Aseptically add the Hepatocyte Differentiation Serum-Free Supplement, growth factors and the Dexamethasone to the basal medium - transfer entire contents of the vial then rinse the vial with basal medium3. Antibiotics are optional but may be added. The cells have been tested in media containing 150ul of Gentamicin in 250ml media
4. Filter through 0.22um filter unit5. Store the complete medium at 4°C in the dark until ready for use
6. Warm the complete medium to room temperature immediately before use
a. Do not leave the complete medium in a water-bath for longer than 5 minutes as this may result in degradation of the medium components Note: Degradation of some components is toxic to immature stem/progenitor cells7. Use complete medium within two weeks8. After 5 days of differentiation remove the differentiation medium and replace with Hepatocyte Maintenance Medium and feed daily.
Storage and Stability:Store the complete medium at 4°C in the dark until ready for use.
