Inhibitory cell surface receptor involved in the regulation of T-cell function during immunity and tolerance. Upon ligand binding, inhibits T-cell effector functions in an antigen-specific manner. Possible cell death inducer, in association with other factors.
Source:Recombinant protein corresponding to aa25-167 from human PD-1, fused to FLAG-Avi-His-Tag at C-terminal, expressed in HEK293 cells.
Molecular Weights:~20kD
Applications:Suitable for use in study of protein binding and screening small molecular inhibotors. Other applications not tested.
Assay Conditions:10nM PD-L1, Biotin labeled was mixed with varying amounts of PD-1, FLAG-Tag in PD-1 Assay Buffer for 1hr at RT. FLAG-acceptor beads (left) or Nickel-acceptor beads (right) were added and incubated for 30min at RT followed by streptavidin-conjugated donor beads for 30min at RT. Alpha counts were read.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
