Test Principle:The microtiter plate provided in this kit has been pre-coated with CCK-8. During the reaction, CCK-8 in the sample or standard competes with a fixed amount of CCK-8 on the solid phase supporter for sites on theBiotinylated Detection Antibody specific to CCK-8. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substratereaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of CCK-8 in the samples is then determined by comparing the O D of the samples to the standard curve.
Storage and Stability:Store unopened kit at 4ºC; store at -20ºC once opened. Store unopened standard at 4°C; once reconstituted, store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Kit is stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
