Test Principle:The microtiter plate provided in this kit has been pre-coated with APLN. During the reaction, APLN in the sample or standard competes with a fixed amount of APLN on the solid phase supporter for sites on theBiotinylated Detection Antibody specific to APLN. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substratereaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of APLN in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Storage and Stability:Store unopened kit at 4ºC; store at -20ºC once opened. Store unopened standard at 4°C; once reconstituted, store at 4°C for up to 12 hours or at -20°C for up to 48 hours. Store other components at 4°C. Kit is stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
