TOP2A encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA.
Sample Type:Paraffin sample (breast cancer)
Intended Use:FISH
Probe Name:TOP2A/CEP17
Fluorophore:Orange/Green
Clinical Significance:1. Determine the prognosis of breast cancer, TOP2A gene abnormalities in breast cancer patients predicts a shorter recurrence-free survival (RFS)
2. Guide clinical medication. Patients treated with TOP2A abnormalities have 65% lower risk of relapse using the CEF regimen than CM regimen, and the risk of death could be reduced by 67%; While TOP2A normal patients using CEF and CMF regimens’ curative effect is similar.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
