Lactate dehydrogenase (LDH) exists in human tissues and organs. LDH is an important enzyme in the energy metabolism of body. The change of LDH quality and quantity will directly affect the body energy metabolism. When the body tissues and organs get diseased, LDH in the tissues and organs will change, and LDH in blood will change too. Increased LDH is mainly found in the acute myocardial infarction, virus hepatitis, cirrhosis of the liver, pulmonary infarction, some malignant tumors, skeletal myopathy, nucleated red blood cell of bone marrow failure (invalid hematopoietic), leukemia, especially acute lymphoblastic leukemia, malignant anemia. Patients with chronic kidney disease of uremia have normal LDH in serum. After dialysis treatment, the LDH activity will increase, which is related to the elimination of LDH inhibitor urea and the oxalate in serum.
Sample Type:Tissues, serum, plasma, culture cells, cell culture supernatants, cerebrospinal fluid and other samples.
Intended Use:This kit can be used to measure LDH activity in tissues, serum, plasma, culture cells, cell culture supernatants, cerebrospinal fluid and other samples.
Test Principle:Lactic acid → LDH → PyruvateIncubate at 37°C, alkaline Pyruvate + 2,4 − dinitrophenylhydrazine →Pyruvic acid dinitrophenylhydrazine (reddish brown). The activity of LDH can be measured according to Beer's law.
Kit Components (see Protocol for final kit quantities):Reagent 1,Substrate buffer, 5 mLx1 vialCoenzyme I, Powderx1 vial
Reagent 2,Preparation of coenzyme I application solution: Add 1.3 mL of double distilled water to each vial of coenzyme powder. The solution can be kept in frozen for 2 weeks. If needed to be used for several times, sub packaging is recommended.
Reagent 4,Preparation of NaOH solution (0.4mol/L): Prepare the needed amount of 0.4mol/L of NaOH solution by adding double distilled water to the NaOH stock solution.
Reagent 5,Pyruvate standard solution, 2umol/mL, 1 mLx1 vialPreparation of pyruvate standard application solution (0.2umol/mL): Dillute 2umol/mL of pyruvate standard solution with double distilled water. Prepare fresh solution before use.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
