Glutathione reductase (GR) is an enzyme, Glutathione reductase catalyzes the reduction of glutathione disulfide (GSSG) to the sulfhydryl form glutathione (GSH), which is a critical molecule in resisting oxidative stress and maintaining the reducing environment of the cell. Glutathione reductase functions as dimeric disulfide oxidoreductase and utilizes an FAD prosthetic group and NADPH to reduce one molar equivalent of GSSG to two molar equivalents of GSH.
Sample Type:Tissue homogenates, cell lysates
Intended Use:This kit is can be used for detection of GR content in tissue homogenates, cell lysates and other biological fluids.
Test Principle:Oxidized glutathione (GSSG) can be reduced to GSH under the catalysis of GR. GSH react with substrate DTNB to produce yellow TNB and GSSG. The production of TNB can be measured by absorbance at 412nm (A412).
Kit Components (see Protocol for final kit quantities):GR Test Buffer, 50 mL Sample Diluent, 50 mLNADPH, 5 mgOxidized glutathione(GSSG), 14.2mgDTNB, 4.5mgDMSO, 1.5mLManual, 1xcopy
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
