Gamma-glutamylcysteine Synthetase (Gamma-GCS) is the rate-limiting enzyme in the synthesis of glutathione (GSH). Intracellular Gamma-GCS activity is increased with the acceleration of GSH synthesis, thereby increasing the concentration of intracellular and extracellular GSH. GSH is an important antioxidant in the body. GSH plays a significant role in maintenance of airway epithelial cell integrity, resistance to lung injury and inflammation and antioxidant damage. The activity of Gamma-GCS and the concentration of GSH (6.1~17.5nmol/mg lung tissue) are both high in the lung. The concentration of GSH in the lining of respiratory tract epithelium (200~400umol/L) is 100 times higher than the one in the plasma. Thus GSH plays an essential role in respiratory inflammation and allergic reactions; and GSH has great research significance in the treatment and prognosis of cardiovascular disease and tumor.
