Test Principle:N-acetyl-beta-D-glucosamidase (NAG) is an acid hydrolase in lysosome and exist wildly in various tissues, organs, body fluid, red blood cells, leukocytes and blood platelets. Substrate hydrolyze under the presence of NAG and release free paranitrophenol. Adding alkaline solution stops the reaction and triggers the color development of paranitrophenol. Active unit of the enzyme can be calculated by measuring the absorbance at 400 nm wavelength.
Kit Components (see Protocol for final kit quantities):Reagent 1,Solution, 40 mLx1 vial
Reagent 2,Substrate, powderx1 vial
Reagent 3,Stop solution, 60 mLx2 vial
Reagent 4,Liquid, 6 mLx1 vial
Reagent 5, 2 mLx1 vial:Paranitrophenol standard stock solution, 3mmol/L
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
