Intended Use:This kit can be used for quantitative determination of NEFA in serum, plasma, tissue homogenate, cells (or cell supernatant).
Test Principle:Glutamic-pyruvic transaminase (Alanine aminotransferase /ALT /GPT) can react with the substrate consisted of alanine and alpha-ketoglutaric acid in pH7.4 solution at 37°C, and then produce alanine and glutamic acid. Add 2,4-dinitrophenylhydrazine (DNPH) hydrochloric acid solution after 30 min to stop the reaction. Then the addition reaction occurs between DNPH and the carbonyl in ketonic acid and produce pyruvic acid hydrazine. The phenylhydrazone can show brown color in alkaline condition. Measure the OD values at 505nm and calculate the enzyme activity.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
