Sphingosine kinase 1 is an enzyme that in humans is encoded by the SPHK1 gene. The protein encoded by this gene catalyzes the phosphorylation of sphingosine to form sphingosine-1- phosphate (S1P), a lipid mediator with both intra- and extracellular functions. Intracellularly, S1P regulates proliferation and survival, and extracellularly, it is a ligand for cell surface G protein- coupled receptors. This protein, and its product S1P, play a key role in TNF-alpha signaling and the NF-kappa-B activation pathway important in inflammatory, antiapoptotic, and immune processes. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
Intended Use:Sandwich High Sensitivity BioAssay™ ELISA kit for Quantitative Detection of Human SPHK1/Sphingosine Kinase 1.
Sensitivity:<10pg/ml
Range:312pg/ml-20,000pg/ml
Specificity:Recognizes natural and recombinant Human SPHK1/Sphingosine Kinase 1. There is no detectable cross-reactivity with other relevant proteins.
Test Principle:This Human SPHK1/Sphingosine Kinase 1 BioAssay™ ELISA Kit was based on standard sandwich enzyme- linked immune-sorbent assay technology. A specific for SPHK1/Sphingosine Kinase 1 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO; Immunogen sequence: M1-L384) and test samples are added to the wells, a biotinylated detection specific for SPHK1/Sphingosine Kinase 1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human SPHK1/Sphingosine Kinase 1 amount of sample captured in plate.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
