Osteoglycin (also called mimecan) is a protein that in humans is encoded by the OGN gene. This gene encodes a protein which induces ectopic bone formation in conjunction with transforming growth factor beta. This protein is a small proteoglycan which contains tandem leucine-rich repeats (LRR). The level of expression of this gene has been correlated with enlarged hearts and more specifically left ventricular hypertrophy. Mimecan belongs to a family of small leucine-rich proteoglycans (SLRPs). The mimecan gene was expressed at a moderate level in the mouse pituitary gland by Northern blot analysis. Expression of mimecan mRNA and protein is also observed in the human anterior pituitary gland. The OGN gene was mapped to chromosome 9q22.

Test Principle:This Mouse Mimecan BioAssay™ ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A a monoclonal antibody from rat specific for Mimecan has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: A20-F298) and test samples are added to the wells, a biotinylated detection a biotinylated polyclonal antibody from goat specific for Mimecan is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse Mimecan amount of sample captured in plate.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
