HMGCL, also known as hydroxymethylglutaryl-CoA lyase, mitochondrial isoform 1, is a mitochondrial matrix protein that belongs to the HMG-CoA lyase family. It is a mitochondrial enzyme that catalyzes the final step of leucine degradation and plays a key role in ketone body formation. Multiple isoforms of the proteins are known due to alternative splicing. The major isoform (isoform 1) is most highly expressed in the liver whereas isoform 2 is found in energy-demanding tissues including the brain, heart, and skeletal muscle.
