Membrane-associated arginine methyltransferase that can both catalyze the formation of omega-N monomethylarginine (MMA) and asymmetrical dimethylarginine (aDMA). Able to mono- and dimethylate EWS protein; however its precise role toward EWS remains unclear as it still interacts with fully methylated EWS.
Source:Recombinant protein corresponding to aa61-394 from human PRMT8, fused to His-FLAG-Tag at N-terminal, expressed in Sf9 cells using a Baculovirus expression system.
Molecular Weight:~40.8kD
Specific Activity:~2.22pmol/min/ug
Assay Conditions:50ul reaction mix (20mM phosphate buffer pH 7.4, 20uM S-adenosyl methionine, and 1-5ng methyltransferase PRMT8) added to the wells coated with the substrate. Incubate at RT for 1 hr. Then, add secondary HRP-labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminesence.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
