Specificity:Recognizes human VCAM-1. There is no detectable cross-reactivity with any of the following tested proteins: IL-1, IL-2, IL-3, IL-4, IL-5, IL-6, IL-8, IL-10, IL-12, TNF, VEGF, TGF or IFN.
Test Principle:This assay employs the Sandwich immunoassay technique. An anti-hVCAM-1 monoclonal coating antibody is adsorbed onto microwells. VCAM-1 present in the sample or standard binds to antibodies adsorbed to the microwells. Following incubation unbound sample or standard are removed during a wash step. A Biotinylated anti-hVCAM-1 antibody is added and binds to VCAM-1 captured by the first antibody. Following incubation unbound Biotinylated anti-hVCAM-1 antibody is removed during a wash step. A Streptavidin-HRP is added and binds to Biotinylated anti-hVCAM-1 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step. A colored product is formed in proportion to the amount of VCAM-1 present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450nm. A standard curve is prepared from seven VCAM-1 standard dilutions and VCAM-1 sample concentration determined.
Storage and Stability:Store *361424B powder at 4°C Aliquot and store reconstituted standard at -70°C. Store all other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
