Interleukin 10 (IL-10), originally known as cytokine synthesis inhibitory factor (CSIF), is a pleiotropic cytokine with a myriad of immunomodulatory effects on a variety of cell types. On activated B cells, IL-10 can induce the formation of plasma cells and the secretion of IgG or, in the presence of TGF-beta1 and/or IL-4, the secretion of IgA. On macrophages, IL-10 is known to downregulate IL-1, TNF-alpha and IL-6 production. On dendritic cells, IL-10 has been shown to interfere with antigen-presenting cell (APC) functions by downmodulating the expression of stimulatory and co-stimulatory molecules. IL-10 has also been found to inhibit dendritic cell differentiation from monocytes and to promote monocyte maturation into cytotoxic CD16+ macrophages. Rat IL-10 cDNA encodes a 178 aa residue precursor protein that contains an 18aa signal sequence and a 160aa mature protein. Mature rat IL-10 shares 85% and 74%aa sequence identity with mouse and human IL-10, respectively.

Test Principle:This assay employs the Sandwich immunoassay technique. An anti-r IL-10 monoclonal coating antibody is adsorbed onto microwells. IL-10 present in the sample or standard and Biotinylated anti-IL-10 antibody binds to antibodies adsorbed to the microwells. Following incubation unbound sample or standard and Biotinylated anti-IL-10 antibody are removed during a wash step. A Streptavidin-HRP is added and binds to Biotinylated anti-IL-10 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step. A colored product is formed in proportion to the amount of IL-10 present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450nm. A standard curve is prepared from seven IL-10 standard dilutions and IL-10 sample concentration determined.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
