The interleukin 1 (IL-1) family of proteins consists of IL-1alpha, IL-1beta and the IL-1 receptor antagonist. IL-1alpha and IL-1beta bind to the same cell surface receptors and share biological functions. With the exception of skin keratinocytes, some epithelial cells, and certain cells of the central nervous system, IL-1 is not produced by unstimulated cells of healthy individuals. However, in response to stimuli such as those produced by inflammatory agents, infections, or microbial endotoxins, a dramatic increase in the production of IL-1 by macrophages and various other cell types is seen. IL-1 possesses a wide variety of biological activities and plays a central role in mediating immune and inflammatory responses. Although normal production of IL-1 is obviously critical for initiation of normal host responses to injury and infection, inappropriate or prolonged production of IL-1 has been implicated as playing a role in the production of a variety of pathological conditions including sepsis, rheumatoid arthritis, inflammatory bowel disease, acute and chronic myelogenous leukemia, insulin-dependent diabetes mellitus, and atherosclerosis.

Test Principle:This assay employs the Sandwich immunoassay technique. An anti-m IL-1beta monoclonal coating antibody is adsorbed onto microwells. IL-1beta present in the sample or standard and Biotinylated anti IL-1beta antibody binds to antibodies adsorbed to the microwells. Following incubation unbound sample or standard and Biotinylated anti IL-1beta antibody are removed during a wash step. A Streptavidin-HRP is added and binds to Biotinylated anti IL-1beta antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step. A colored product is formed in proportion to the amount of IL-1beta present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450nm. A standard curve is prepared from seven IL-1beta standard dilutions and IL-1beta sample concentration determined.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
