Ficolin-2, which was initially identified as L-ficolin, is a protein that in humans is encoded by the FCN2 gene. The product of this gene belongs to the ficolin family of proteins. This family is characterized by the presence of a leader peptide, a short N-terminal segment, followed by a collagen-like region, and a C-terminal fibrinogen-like domain. It is mapped to 9q34.3. This gene is predominantly expressed in the liver, and has been shown to have carbohydrate binding and opsonic activities. Serum concentration of ficolin-2 varies considerably in healthy individuals. FCN2 promoter polymorphisms were associated with marked changes in theficolin-2 serum concentration.
Sample Type:Cell culture supernates, serum and plasma (heparin)
Intended Use:For quantitative detection of human Ficolin-2 in cell culture supernates, serum and plasma(heparin).
Sensitivity:<20pg/ml
Range:0.78-50ng/ml
Specificity:Natural and recombinant human Ficolin-2
Test Principle:This human Ficolin-2 BioAssay™ ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Ficolin-2 has been precoated onto 96-well plates. Standards(NSO, L26-A313) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ficolin-2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Ficolin-2 amount of sample captured in plate.
