Complement factor H (CFH), originally known as beta-1H globulin, is a serum glycoprotein that regulates the function of the alternativecomplement pathway in fluid phase and on cellular surfaces. It binds to C3b, accelerates the decay of the alternative pathway convertase C3bBb, and also acts as a cofactor for complement factor I, another C3b inhibitor. The CFH gene is located on chromosome 1q32-q32.1 within a cluster of genes encoding the regulatory complement components of the activation of C3 (RCA for 'regulators of complement activation'). This gene cluster includes decay-accelerating factor (DAF), C4-binding protein (C4BPA and C4BPB), and the factor H-related genes CFHR1, CFHR2, CFHR3, CFHR4, and CFHR5, among others. The gene family has arisen by multiple duplication events.
Sample Type:Cell culture supernates, serum and plasma (heparin, EDTA)
Intended Use:For quantitative detection of mouse CFH in cell culture supernates, serum and plasma (heparin, EDTA).
Sensitivity:<20pg/ml
Range:0.78-50ng/ml
Specificity:Natural and recombinant mouse CFH
Test Principle:This mouse CFH BioAssay™ ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CFH has been precoated onto 96-well plates. Standards (NSO, S875-V1252) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CFH is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CFH amount of sample captured in plate.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
