Complement factor D(CFD), also called ADIPSIN or FACTOR D, is a protein which in humans is encoded by the CFD gene. The protein encoded by this gene belongs to the trypsin family of peptidases. It is mapped to 19p13.3. The encoded protein is a component of the alternative complement pathway which is best known for its role in humoral suppression of infectious agents. This protein is also a serine protease that is secreted by adipocytes into the bloodstream. Finally, the encoded protein has a high level of expression in fat, suggesting a role for adipose tissue in immune system biology. Factor D is a serine protease that stimulates glucose transport for triglyceride accumulation in fats cells and inhibits lipolysis.

Test Principle:This human CFD BioAssay™ ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CFD has been precoated onto 96-well plates. Standards(NSO, I26-A253) and test samples are added to the wells; a biotinylated detection polyclonal antibody from goat specific for CFD is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CFD amount of sample captured in plate.
