• This protein is a member of the TNF-receptor superfamily. This protein is an osteoblast-secreted decoy receptor that functions as a negative regulator of bone resorption. This protein specifically binds to its ligand, osteoprotegerin ligand, both of which are key extracellular regulators of osteoclast development. Studies of the mouse counterpart also suggest that this protein and its ligand play a role in lymph-node organogenesis and vascular calcification. Alternatively spliced transcript variants of this gene have been reported, but their full length nature has not been determined.

• Specificity:This assay has high sensitivity and excellent specificity for detection of mouse OPG. No significant cross-reactivity or interference between mouse OPG and analogues was observed.
• Test Principle:This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for OPG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any OPG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for OPG is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of OPG bound in the initial step. The color development is stopped and the intensity of the color is measured.
