• Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils.

• Specificity:This assay has high sensitivity and excellent specificity for detection of human MPO. No significant cross-reactivity or interference between human MPO and analogues was observed.
• Test Principle:This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MPO has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MPO present is bound by the immobilized antibody. After removing any unbound substances, a Horseradish Peroxidase (HRP)-conjugated antibody specific for MPO is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPO bound in the initial step. The color development is stopped and the intensity of the color is measured.
