• This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. This gene is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. In addition, aberrant expression of this protein is observed in cervical adenocarcinomas. Mutations in this gene have been associated with congential hypotrichosis with juvenile macular dystrophy.

• Specificity:This assay has high sensitivity and excellent specificity for detection of human P-cad. No significant cross-reactivity or interference between human P-cad and analogues was observed.
• Test Principle:This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for P-cad has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any P-cad present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for P-cad is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of P-cad bound in the initial step. The color development is stopped and the intensity of the color is measured.

• Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
