• This gene encodes a protease inhibitor that regulates the tissue factor (TF)-dependent pathway of blood coagulation. The coagulation process initiates with the formation of a factor VIIa-TF complex, which proteolytically activates additional proteases (factors IX and X) and ultimately leads to the formation of a fibrin clot. The product of this gene inhibits the activated factor X and VIIa-TF proteases in an autoregulatory loop. The encoded protein is glycosylated and predominantly found in the vascular endothelium and plasma in both free forms and complexed with plasma lipoproteins. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been confirmed.

• Specificity:This assay has high sensitivity and excellent specificity for detection of human TFPI. No significant cross-reactivity or interference between human TFPI and analogues was observed.
• Test Principle:This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TFPI has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TFPI present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TFPI is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TFPI bound in the initial step. The color development is stopped and the intensity of the color is measured.
