特徴

• The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and kininogens. This gene encodes a stefin that functions as an intracellular thiol protease inhibitor. The protein is able to form a dimer stabilized by noncovalent forces, inhibiting papain and cathepsins l, h and b. The protein is thought to play a role in protecting against the proteases leaking from lysosomes. Evidence indicates that mutations in this gene are responsible for the primary defects in patients with progressive myoclonic epilepsy (EPM1).

• Specificity:This assay has high sensitivity and excellent specificity for detection of human cystatin-B. No significant cross-reactivity or interference between human cystatin-B and analogues was observed.
• Test Principle:This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for cystatin-B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any cystatin-B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for cystatin-B is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of cystatin-B bound in the initial step. The color development is stopped and the intensity of the color is measured.
