• Specificity:This assay has high sensitivity and excellent specificity for detection of mouse Cortisol. No significant cross-reactivity or interference between mouse Cortisol and analogues was observed
• Note: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between mouse Cortisol and all the analogues. Therefore, cross-reaction may still exist.
• Principle of the Assay:This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate has been pre-coated with an antigen. Standards andsamples are added to the appropriate microtiter plate wells with antibody specific for Corisol and goat anti-rabbit (HRP) antibody. The competitive inhibition reaction is launche between the pre-coated cortisol and cortisol in the samples. A substrate solution is added to the wells and the color develops opposite to the amount of cortisol in the sample. The color development is stopped and the intensity of color is measured.

• Storage and Stability:Store kit at 4°C. The unopended kit is stable for 6 months after receipt. Once opened, the kit is stable up to 4 weeks at 4°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap
• Assay Summary:1. Prepare reagents and samples as instructed
• 2. Set a Blank well without any solution3. Add 50ul Standard or sample to each well
• 4. Add 50ul Antidody, 1X to each well (do not add any to the blank well)
• 5. Incubate for 40 minutes at 37°C
• 6. Aspirate and wash 3 times
• 7. Add 100ul HRP Conjugate, 1X to each well (do not add any to the blank well)
• 8. Incubate for 30 minutes at 37°C
• 9. Aspirate and wash 5 times
• 10. Add 90ul TMB Substrate to each well. Incubate for 20 minutes at 37°C. Protect from light11. Add 50ul Stop Solution to each well. Read at 450nm within 5 minutes.
