• NMNAT2 belongs to the nicotinamide mononucleotide adenylyltransferase (NMNAT) enzyme family, members of which catalyze an essential step in NAD (NADP) biosynthetic pathway. unlike the other human family member, which is localized to the nucleus, and is ubiquitously expressed; this enzyme is cytoplasmic, and is predominantly expressed in the brain. Two transcript variants encoding different isoforms have been found for this gene.

• Specific Activity:~500pmol/min/ug
• Unit Definition:One unit will convert 1.0pmole of beta-NADH/minute to beta-NAD at pH 8.0 at 37°C.
• Activity Assay:1. Prepare a 200ul reaction buffer into a suitable container: The final concentrations are 20mM Tris-HCl, pH 8.0, 10mM MgCl2, 1mM ATP, 1% ethanol, 1mM beta-NMN, 100mM sodium chloride, 6 unit ADH
• 2. Equilibrate to 37°C and monitor at A355nm until the value is constant using a spectrophotometer
• 3. Add 20ul of recombinant NMNAT2 protein with various concentrations (5ug, 2.5ug, 1.25ug) in 180ul reaction buffer
• 4. Record the increase at A355nm for 15 minutes.
• Applications:Suitable for use in SDS-PAGE. Other applications not tested.
• Recommended Dilution:Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
