A cell-permeable bis-nitrobenzylidene-piperidinone compound that acts as a potent, reversible and selective inhibitor of 19S regulatory-particle-associated deubiquitylating enzymes (DUBs) UCH-L5 and USP-14 (IC50=2.1uM against Ub-AMC substrate), with no effect on UCH-L1, UCH-L3, USP2, USP7, USP8 and BAP1 and on the proteasomal proteolytic activities. Shown to prevent proteasomal degradation (IC50=0.8uM in MelJuSo-UbG76V-YFP cells), and cause accumulation of higher molecular weights polyubiquitinated proteins in HCT-116 cells. Preferentially induces cell-cycle arrest and apoptosis in several colon cancer and in CNS cells, and effectively suppresses tumor growth in various mouse model (5mg/kg, i.p.).
