特徴

• Flagellin is the subunit protein which polymerizes to form the filaments of bacterial flagella. It activates the innate immune system through the receptor Toll-like Receptor 5 (TLR5) or the intracellular NLRC4 protein. The Flagellin (TLR5 Mutant) cannot be detected by human and mouse TLR5. It has not been tested on NLRC4.
• Source:Recombinant protein corresponding to native flagellin [FliC] from Salmonella enterica Serovar Typhimurium strain ATCC14028, aa1-495, contains a substitution of aa90-97 required for TLR5 signaling by a motif from a non-signaling flagellin, fused to His-tag at N-terminal, expressed in E. coli
• Biological Activity:Does not activate TLR5 in human epithelial cell assays based on NF-kB luciferase fusions, in vivo on expression of CCL20 or IL-6. It can be used as negative control for TLR5 signaling assays.
• Endotoxin:<0.01EU/ug purified protein (LAL test; Lonza)
• Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
