Human proprotein convertase subtilisin/kexin type 9 (PCSK9), also known as FH3, HCHOLA3, and PC9. PCSK9 is autocleaved to the ~14kD prodomain (aa31-152) and the ~58kD mature form (aa153-692(end)), which run at higher apparent MW by SDS-PAGE.
Recombinant protein corresponding to aa31-692 from human PCSK9, fused to His-tag, expressed in HEK293 cell expression system.
Assay Conditions:Coated plate with 100 ng/well of PCSK9-his (172343) in 50ul (in PBS) (overnight at 4°C. Initiated binding reaction with addition of various concentrations of LDLR- Biotin in 50mM HEPES pH7.4, 50mM sodium chloride, 0.2mM calcium chloride, 0.5% Tween-20, 0.1% BSA or 50mM MES pH6, 50mM sodium chloride 0.2mM calcium chloride, 0.5% Tween-20, 0.1% BSA. Incubated for 2h at RT. Detected binding with Strep-HRP generated luminescence. No specific binding detected in HEPES pH7.4 buffer formulation
Applications:Suitable for use in studying protein binding and screening small molecules for drug discovery. Other applications not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
