Human phosphoglycerate dehydrogenase (PGDH), also known as PDG, amino-acids 1 – 533 (end) (GenBank Accession No. NM_006623) with C-terminal His tag, MW = 57.5kD, expressed in an E. coli expression system.
Source:Recombinant protein corresponding to aa1-533 from human PHGDH at C-terminal, fused to His-tag expressed in E.coli.
Molecular Weight:~57.5kD
Specific Activity:80 pmol/min/ug
Assay Conditions:The reaction mixture contained 100mM Tris-HCl, pH 8.8, 400mM NaCl, 0.2mM DTT, 2.5mM 3-phospho-D-glyceric acid (Sigma P8877), 1mM NAD, and various amount of PHGDH in a final volume of 50ul. Reactions were initiated with the addition of PHGDH, and the increase in NADH fluorescence (Ex340/Em460 nm) was continuously monitored for 10 min at room temperature.
Application:Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap
