Assay Conditions:Enzyme reaction was done in a buffer (50mM Tris, pH 7.4, 50mM NaCl, 10mM CaCl2, and 2mM DTT) containing PADI4 and 10mM BAEE as a substrate for 25 min at 37°C. Amount of ammonia produced was measured and used for specific activity calculation.
Application:Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
