Assay Conditions:IDH reductive activity was measured in 200ul reaction containing 25mM Tris (pH 7.4), 150 mM sodium chloride, 10mM MgCl2, 0.03% BSA, 1mM alpha-Ketoglutarate, 10uM NADPH and IDH. Depletion of NADPH was monitored continuously at 340nm for 20 minutes. Molar extinction coefficient of NADPH is 6,200 M-1cm-1
Application:Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested.
Recommended Dilutions:Optimal dilutions to be determined by the researcher.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
