• IMBP plates are ideally suited for applications requiring the detection, isolation and/or characterization of rare, minority contaminating sequences. Because amplification will amplify both minority and majority sequences, denaturing and annealing will result in virtually all of the minority strands pairing with majority strands, thus producing mismatch containing DNA. These heteroduplexes can then be isolated by the IMBP method. It is possible to release bound heteroduplexes from IMBP and subsequently to amplify, sequence or clone them. For example, we have been successful in enriching for DNA fragments that were initially 1% or less of the original DNA mix. Following enrichment, these minority sequences were detected as clear heterozygotes by both manual and automated sequencing. As with all IMBP allications, good results depend on good input DNA (misincorporation errors during amplification wil also produce heteroduplexes which can be bound by IMP/BP and increase the background).
